Description of VisuLize™ Buffer Pak
The VisuLize™ Buffer Pak consists of assay plates, general purpose buffers and materials for the preparation and performance of ELISA assays. This Buffer Pak provides sufficient reagents for 5 x 96 well assays. The components of the VisuLize™ Buffer Pak can be used in the development of in-house immunoassays and are especially suited for use with a number of Affinity Biologicals’ matched-pair antibody sets for EIA, including the following:

Some of the EIA sets listed may not require all the components provided in the VisuLize™ Buffer Pak. Please refer to specific EIA Instructions for Use for the appropriate buffers and assay conditions.
This kit is intended for use by personnel trained in laboratory procedures and precautions for the use of chemicals.
The OPD substrate (o-Pheylenediamine.2HCl) is toxic therefore precautions should be taken to avoid direct contact. The use of gloves and safety glasses is recommended.
The disposal of waste materials must be carried out according to current local regulations.

Supplied Materials:
Item 1. 5 x 96 well microtiter plates; each plate consists of 6 x 16 well strips and frame
Item 2. 5 Adhesive Plate sealers
Item 3. 1 bottle containing 60 mL Coating Buffer (50 mM Carbonate, pH 9.6, plus preservative); to be used in coating of antibodies to microtiter plates
Item 4. 1 bottle containing 80 mL Blocking Buffer (1% BSA in PBS, pH 7.4, plus preservative); to be used as blocking reagent in ELISA assays
Item 5. 2 bottles, each containing 125 mL 20X PBS-Tween (2% (v/v) Tween-20 in PBS, pH 7.4, plus preservative); to be used as wash buffer and/or sample diluent, depending on assay requirements
Item 6. 5 tubes, each containing 10 mL 5X HBS-BSA-T20 (0.5% (v/v Tween-20, 5% BSA in HBS, pH 7.2, plus preservative); to be used as sample diluent and/or detecting antibody diluent, depending on assay requirements
Item 7. 1 bottle containing 70 mL Substrate Buffer (Citrate-phosphate buffer, pH 5.0 plus preservative); to be used as buffer for OPD substrate
Item 8. 5 x 5mg OPD tablets (o-Pheylenediamine.2HCl)
Materials Required but not Provided
Reagent grade water
Single channel and multi-channel pipettes
Pipette tips
2.5 M H2SO4
30% H2O2
Microplate washer (optional) and reader
Reagent Preparation:
Prior to use, allow reagents to warm to room temperature.
Item 1. (6 x 16 well strips with frame): Prior to use, ensure desired number of strips are assembled in plate frame. Unused strips can be stored in bag.
Item 5. (20X PBS-Tween): Allow bottle to warm to room temperature before use. Ensure any crystals that may have formed are dissolved before proceeding. If necessary the vial can be warmed to 37oC until all crystals have dissolved. Dilute the concentrate 1/20 before use. For every full 96-well plate assay, add 50 mL of 20X PBS-Tween to 950 mL reagent grade water and mix. Stability after dilution is 1 week at 2–8oC.
Item 6. (5X HBS-BSA-T20): Allow vial to warm to room temperature before use. Dilute the concentrate 1/5 before use. For every full 96-well plate assay, add 10 mL of 5X HBS-BSA-T20 to 40 mL reagent grade water and mix. Stability after dilution is 1 week at 2–8oC.
Item 8. (5m OPD tablet): Make up immediately before use. Dissolve 5mg OPD tablet in 12 ml substrate buffer (Item 7) then add 12 μl 30% H2O2. Do not store.
Items 2-4 and 7 are supplied ready to use.

Intact kits and un-reconstituted reagents are stable until the expiration date stated on the box and individual reagent labels when stored at 2–8°C.
